Effect of the addition of methyl-β-cyclodextrin in sperm freezing media for dairy cattle

The effect of the addition of cholesterol-loaded methyl -β-cyclodextrin in freezing media was investigated in order to evaluate post-thaw sperm quality. Prior to the evaluation, the pre-dilution was carried out 1: 1 dividing it into 3 aliquots, for the inclusion of CLC (T0: 0 mg, T1: 1.5 mg, T2: 3.0 mg) which were incubated at 37ºC for 15 minutes, after this time the final dilution was made by immediately soaking them into 0.5 ml straws, taking them to the automatic freezer, gradually lowering the temperature to -196ºC and storing them in the liquid nitrogen tank, the straws were defrosted every 7 days to their respective valuation. For the statistical analysis, a Completely Randomized Design was used, obtaining significant differences P (<0.01) for viability, motility, vitality and integrity of the membrane with means of 56.00%; 63.50%; 75.36%; 60.08% respectively belonging to T1, in terms of morphoanomalies, no statistical differences were found, however T2 presented a higher percentage with a mean of 5.40%; the best level of use being T1, recommending the addition of 1.5 mg CLC in cryopreservation media since, according to the results obtained, better reproductive parameters are expected to be obtained.
